The blades and knifes consumables are for use with microtomes and cryostats. Microtomes and cryostats are devices used to produce consistent thickness slices of tissue samples, they are then mounted on glass slides, stained and typically examined by microscopy. Microtomes for light microscopy typically use disposable blades, to maintain the quality of the cut and thus the section quality. Therefore they must be changed regularly. The frequency of changing depends on the number of sections cut, hardness of the tissue and appropriate use.

Dehydrant Alcohol product lines are specifically designed for tissue processing and staining of histological and cytological specimens. The products are a clear, colorless blend of ethyl, isopropyl, and methyl alcohol that in some cases have trace amounts of other denatured materials. All Dehydrant Alcohol products are non-controlled substances and do not require record keeping. The formulations are readily miscible with water to form any desired grade of alcohol and can be used with all clearing reagents. The Dehydrant Alcohol line can be used with both open and closed tissue processors. Protective gloves should be worn and work should be performed in a ventilated area. Dehydrant Alcohol is a flammable product and must be stored in a flammable fire cabinet. The Dehydrant Alcohol product line can also be used for hydrating and dehydrating tissue sections during staining. They can be used with all special stains that require dehydration, as solvents for preparing special stains, and immunohistochemical procedures that are compatible with alcoholic dehydrants. The Dehydrant Alcohol product line can be substituted in place of any alcohol you are presently using for hematoxylin and eosin-y staining. They are compatible with all manual procedures and automatic stainers.

Formal-Fixx is a variation of neutral buffered formalin with a differing buffering system. This fixative is designed especially for use with the Cytoblock processing gel. The phosphate-free buffering system prevents the formation of free formic acid, and its associated artifacts, formalin pigment. The osmolality of these fixatives allows them to be used not only for light microscopy but for electron microscopic studies as well. Zinc Formal-Fixx Concentrate is the same formulation as Formal-Fixx Concentrate but includes the mineral zinc to enhance the nuclear detail of the specimen.

The Paraffin line is specially designed for tissue processing and embedding of histological and cytological specimens. The product is a white solid at room temperature and a clear, liquid when molten. The paraffin can be used with both open and closed tissue processors. It is also compatible with all embedding centers and manual embedding techniques. Paraffin is used in tissue processing during the infiltration steps. Melting points and polymer content differ between formulations. Choosing which type of paraffin may depend on the tissue density or personal preference of the histotechnician. Softer kinds of paraffin with less polymer tend to infiltrate tissue slightly quicker during processing but yield a softer block during embedding. Softer kinds of paraffin better lend themselves to thicker sections during microtomy. Harder kinds of paraffin with a higher polymer content usually take a little more time to infiltrate the tissue during processing but yield harder blocks that allow thinner sectioning during microtomy. Harder tissue specimens are better supported with a harder paraffin block.

Clear-Rite, Citrus Clearing Solvent, Xylene Substitute, Xylene, and Toluene are a group of solvents known to act as clearing reagents in the histology laboratory for tissue processing and staining. These clearing reagents are used to “clear” tissue or make it permeable to paraffin in tissue processing. In staining, Clearants are used after alcohol which dehydrates the tissue to enable the slide to be cover slipped with a resinous mounting media.

Fixation is the initial and most important step of histology specimen preparation in pathology. 10% Neutral Buffered Formalin is the most used fixative in the histology laboratory, but in certain circumstances, other fixatives may be desired according to tissue type or subsequent pretreatment or staining technique. Pen-Fix is an alcoholic formalin variation with the addition of a proprietary buffer salt.

Signature Series Flex is a patented dehydrating reagent specifically designed for processing and staining of histological and cytological tissue specimens. Flex is a clear, colorless blend of isopropyl and methyl alcohol. It is a non-controlled substance and does not require record keeping. Flex is available in concentrations of 80%, 95% and 100%. It is readily miscible with water to form any desired grade of alcohol. Flex is miscible with all clearing reagents. Studies have indicated that Flex does not over-dehydrate small tissue specimens as rapidly as ethyl and isopropyl alcohol, when used in accordance with the processing guidelines developed for Flex. The absence of over-dehydration during tissue processing allows for consistent results with both large and small tissue specimens. Flex can be used in tissue processing, staining and as a dye or stain solvent in place of ethyl alcohol. Flex can be used in all special stains that require dehydration in ethyl alcohol and in immunohistochemical reactions that are compatible with alcoholic dehydrants. Flex has been proven to produce brilliant hematoxylin and eosin stains, with crisp nuclear detail when used within designated guidelines. Flex is compatible with both open and closed tissue processors. Flex is a flammable product and must be stored in a flammable fire cabinet.

The Fast Flex Tissue Processing System was designed to accommodate a wide range of tissue processing demands. These reagents, when used together with the STP 420ES or Pathcentre tissue processors, will significantly reduce tissue processing times and produce very high-quality results. The Fast Flex Tissue Processing System provides optimal tissue processing and staining results based on the patented rotational technology in the main chamber of the STP 420ES and the chemically enhanced Fast Flex reagents. The Fast Flex Tissue Processing System also produces excellent tissue and staining results when used with the Pathcentre tissue processor. Tissue/chemical interactions, reactions, and diffusion rates are accelerated, and depending upon tissue thickness, fatty tissue such as breast, bowel, and lymph node process very efficiently within this system. Our experience has additionally shown that bloody tissues also process well, especially when Holding Solution is utilized in the grossing room. Traditional hematoxylin and eosin staining demonstrates well-delineated nuclear chromatin, as well as superb cytoplasmic contrast.

Clarifier™ 2 is an aqueous glacial acetic acid rinse designed to eliminate background staining sometimes caused by excessive adhesives in the water bath such as gelatin. Clarifier™ 2 selectively removes hematoxylin staining from excess adhesive without affecting nuclear staining. This product is only effective with progressive staining an should not be used with regressive stains. Clarifier 2 was designed to be used in histology only. Clarifier™ 2 is designed for use after staining with progressive hematoxylins. After tissues have been sufficiently stained with a progressive hematoxylin, slides are removed from the stain and excess hematoxylin is removed by rinsing the sections in water. The sections are then placed into Clarifier™ 2 for 20 seconds to 40 seconds. A 1-minute water rinse should follow as this stops the reaction and rinses excess acid from the tissue section.

Bluing Reagent is intended to be used as an aid in hematoxylin staining for the diagnosis of general pathology specimens. Bluing Reagent is a buffered alkaline rinse that shifts the final hue of hematoxylin from reddish-blue to traditional blue-purple. Unlike ammonia water and lithium carbonate, Bluing Reagent inhibits pH changes that can adversely affect nuclear detail and “crispness”.

Hematoxylin 7211 is a rapid, progressive hematoxylin. The product is a hybrid between classical Harris and Gill formulations: it provides the classic intensity and hues of a Harris hematoxylin and the rapid progressive qualities of a Gill hematoxylin. Differentiation is not required, but if desired Clarifier 1 should be used. Clarifier 1 will eliminate any background staining that may be caused by the use of excess gelatin or other adhesives in the water bath. Clarifier 2 can be used as well; however, a shorter immersion time is required. Hematoxylin 7211 does not have an affinity for acid mucopolysaccharide (mucin) staining, as do some other hematoxylin formulations. It provides consistent, well-delineated nuclear staining results.
